體外研究(In Vitro)
體外研究活性	LB-100 可有效抑制BxPc-3和Panc-1細(xì)胞的生長，IC50值分別為2.3 μM和1.7 μM。BxPc-3、Panc-1和SW1990細(xì)胞經(jīng)LB-100處理后，PP2A活性下降30-50%。LB-100提高細(xì)胞內(nèi)的阿霉素濃度(約為對照組的2.5倍)，并使腫瘤細(xì)胞對阿霉素的細(xì)胞毒性更加敏感。LB-100增加VEGF分泌，從而提高HIF-1α-VEGF介導(dǎo)的血管生成。^[1]
激酶實驗	PP2A 酶活性檢測
激酶實驗	人工培養(yǎng)的胰腺癌細(xì)胞用IC50濃度下的LB-100處理，對每種細(xì)胞系或等體積的載體對照分別處理2小時，然后進(jìn)行PP2A活性測試使用絲氨酸/蘇氨酸磷酸酶試劑盒。細(xì)胞通過超聲波細(xì)胞粉碎機(jī)裂解，PP2A濃度使用絲氨酸/蘇氨酸磷酸酶試劑盒根據(jù)說明測量。每個細(xì)胞系進(jìn)行三個平行試驗。
