            體外研究活性

            Carboplatin作用于一組人類卵巢癌細胞系,包括A2780, SKOV3,和 IGROV-1細胞,抑制細胞增殖,IC50分別為6.1 μM, 12.4 μM 和2.2 μM。[1]

            方法

            MTT實驗: 指數(shù)生長的A2780, SKOV3, IGROV-1和 HX62卵巢癌細胞接種在96孔板中。加入一系列濃度的Carboplatin,再溫育72小時。每組實驗按一式三份進行。 Sulforhodamine B (SRB)實驗:指數(shù)生長的 A2780細胞接種在96孔板中。使用濃度不斷增加的17-AAG 和Carboplatin處理細胞96小時。使用濃度不斷增加的17-AAG或Carboplatin處理細胞 24小時,研究17-AAG 或 Carboplatin處理序列的效果。選中的第一種藥劑處理細胞24小時,確保A2780細胞使用第一種藥劑至少處理雙倍時間(18-24小時)。使用無菌磷酸鹽緩沖液沖洗細胞,然后補充培養(yǎng)基。然后,加入第二種藥劑 (第一次24小時未處理細胞的)或培養(yǎng)基,再處理96小時。所有實驗按一式三份進行。使用中位數(shù)效果分析方法按行之有效的原則分析聯(lián)合研究的效果。使用內(nèi)部電子表格計算聯(lián)用的效果。
