            體外研究活性 RGFP966是作用緩慢的競(jìng)爭(zhēng)性緊密結(jié)合的HDAC抑制劑,抑制HDAC3的IC 50為0.08 μM,而在濃度高達(dá)15 μM時(shí),對(duì)任何其它的HDAC無(wú)有效抑制。[1] 在2種CTCL細(xì)胞系中,RGFP966處理24小時(shí)后免疫印跡分析,結(jié)果表明H3K9/K14,H3K27和H4K5的乙酰化增加,而H3K56ac沒有。RGFP966降低CTCL(皮膚T細(xì)胞淋巴瘤)細(xì)胞生長(zhǎng),是由于細(xì)胞凋亡增加,這是與DNA損傷有關(guān)的和受損的S期相關(guān)聯(lián)。RGFP966導(dǎo)致藥物治療的第一個(gè)小時(shí)內(nèi)顯著減少DNA復(fù)制叉的速度。[2]

            方法 細(xì)胞計(jì)數(shù)并分在T25(Corning)培養(yǎng)瓶中,密度是26105個(gè)細(xì)胞/ mL。然后將細(xì)胞用DMSO或HDIs在0小時(shí)處理一次。100ml的等分試樣從每個(gè)培養(yǎng)瓶中取出,一式三份,在處理0小時(shí),24小時(shí),48小時(shí),和72小時(shí)后取出并分到96孔板中,10毫升阿爾瑪藍(lán)加入到每個(gè)孔中。孵育4小時(shí)后,熒光使用Biotek Synergy MX酶標(biāo)儀測(cè)定。
